Summary

This study investigates Growth/differentiation factor–15 (GDF15), a secreted cytokine strongly linked to dementia risk. Using multiple cohorts, researchers found that plasma GDF15 levels measured in midlife are associated with greater dementia risk over 15- to 25-year follow-up periods, with stronger associations for vascular dementia compared to Alzheimer's disease. Two-sample Mendelian randomization analysis supported GDF15's mechanistic role in Alzheimer's disease and related dementias, suggesting GDF15 may be both a biomarker and a driver of dementia risk.
