Summary
This phase 1, multicentre, randomised, open-label trial (HVTN 140/HPTN 101 part B) evaluated the safety, pharmacokinetics, and neutralisation activity of a combination of three broadly neutralising monoclonal antibodies (PGDM1400LS, VRC07-523LS, and PGT121.414.LS) targeting different HIV-1 epitopes, administered intravenously or subcutaneously in healthy adults without HIV-1 across sites in the USA, Kenya, South Africa, and Zimbabwe. The mAb combination was found to be safe and well tolerated, with no antagonistic pharmacokinetic interactions or loss of neutralisation activity, supporting further evaluation in future efficacy trials for HIV-1 prevention.
