Summary
Researchers developed an exogenous precursor-assisted fungal metabolite (ePAFM) strategy to generate bioactive fungal metabolites, identifying IM502—a meroterpenoid-like compound—as a potent modulator of tumor-associated macrophages (TAMs). IM502 works by inhibiting PI3Kγ and redirecting STAT signaling from STAT3/6 to STAT1/2 dominance, reversing TAM-mediated immunosuppression. This reprogramming of macrophages potently activates anti-tumor immunity, suppresses cancer growth and metastasis, and overcomes resistance to checkpoint blockade immunotherapy, revealing a promising new approach for cancer treatment.
