Laser phase plate enhances cryo-EM resolution for small protein structure determination
By
Holger Müller
Summary
This article presents research demonstrating that a laser phase plate (LPP) installed in a Titan Krios cryo-EM microscope significantly improves the resolution of single-particle reconstruction for small proteins. While previous phase plate designs were unstable and compromised high-resolution signals, the LPP enhances specimen-motion correction and information recovery, overcoming longstanding limitations in cryo-EM imaging of small proteins.

· 3 pulledPhase plates can in principle overcome the poor image contrast in electron cryo–microscopy (cryo-EM) and the resulting limits on the structural reconstruction of small proteins.
However, previous designs have been unstable and compromised the high-resolution signal.
Here, we show that the laser phase plate (LPP), installed in a custom, modern Titan Krios microscope, enhances the resolution in single-particle reconstruction of small proteins by improving specimen-motion correction, recovery of information from t
